Couple of issues here.
1) Environment Variable names are case-sensitive. $DIR and $dir are not the same variable.
2) Your messages appear to be the wrong way round. When we find no files (and the -z condition is satisfied) our message should be "Empty".

Code:
FILE=""
dir="/ann/a1"

if [ -z "$(ls $dir)" ]; then
     echo "$dir is Empty"
else
     echo "$dir is not Empty"
fi

gvfs-monitor-dir - monitor the change of a directory content using the VFS system SYNOPSIS
gvfs-monitor-dir URI DESCRIPTION
