In simple terms it is the unit of measure that changes to the file system are made. Think of the file system as a big grid, where each square has an address. Data is written to this square, but the whole square is used for this, regardless of the size of the data in it. For example, with a 512k block size, your 65 byte file would take up 512k. Modern file system can subdivide the block for more efficient use of space.
