Could you please try following and let me know if this helps you.
Code:
awk '{$2=$2==0?1:$2;$4=$4==0?1:$4} 1' OFS="\t"  Input_file
OR
awk '{gsub("0","1",$2);gsub("0","1",$4)} 1' OFS="\t" Input_file

Output will be as follows.
