The if condition has only the variable name and not it's value.. you could try as
Code:
if [ -s "${filename}" ]; then 
	DIFF=`diff filename filename2`
	if [ "${DIFF}" ]; then
		echo "Hello"
	else
		echo "Another Hello"
	fi
else
	if [ -e "${filename3}" ]
        then		
		echo "Last Hello"
	fi
fi
