For the first character other than spacing characters

It depends on what you want to do with the output.

Without any further information, the obvious choice is:

cut -c1 "$file"

It could be used, for example, to count the frequency of letters that begin words:

file=/usr/share/dict/words
cut -c1 "$file" | sort | uniq -c | sort -n
