Without more information about what you are trying to accomplish, it's hard to post any useful advice. If you are completely unfamiliar with Perl, why do you require the solution to be in Perl in the first place? If you have an existing Perl script which you want to modify, why don't you post (at least part of) the script here for us to see?
Now my aim is to search for the line with number '333' and update at the end of the line that corresponds to this number with the value '|1.6'
before '}'....
I have written the code like this.....
#!/usr/bin/perl -p
`cat tmp | sed \"s/\(\|333\}.*)\\}\$/$1|1.6}/\"`;
But this code is not updating the line with the value 1.6...
What I posted before should work. The shell script you have there now is not Perl code (but might more or less work if you change the first line to #!/bin/sh ... although the cat is bad style, and the backward quotes are needless, if not even harmful).
I added the -i option as you presumably want the file you run it on to actually change. (Many times, you find scripts which will merely print the modified file, and saving the modified contents somewhere would be left as an exercise.)
If you don't have any particular reason to require a Perl solution, it's probably a bad idea to ask for one specifically. As you saw, you got good replies with solutions in other languages anyway, for better or for worse.
