Code:
wc -c filename

Note: each carriage return (new line) counts as a character.

This should help:

Code:
awk -F"R" '{counter+=(NF-1)}END{print counter}' file.txt
20

# 4

Code:
awk 'END{print NR-1}' RS=R infile

# 5

Or
Code:
wc -lc filename | read a b notused && echo $(( $b - $a ))

# 6

And just for the fun of it, the old way:
Code:
fold -1 filename | grep -c "R"

20

And a lateral thought way. Delete everything you are not interested in and count what is left.

Code:
cat filename | tr -cd "R" | wc -c

20

grep -o R file | wc -l

Code:
tr -cd R <file | wc -c
