Then we're back to a permissions error. did you make it world writable or chown it to the user that your web server runs as?

The program runs beautifully from the command line. I think that shows that the permissions are OK.

The PHP script dies w/ an error.

The opposite. It almost exclusively proves a permission error. If the webserver is calling a PHP script, it generally does this as www-data:www-data or some other user.

You were correct - it was the permissions.

Now set as crwxrwxrwx 1 root dialout 4, 64 Oct 27 20:29 /dev/ttyS0
and works perfectly.

The C program hangs frjom the command line.

It has been explained to you -- repeatedly -- that PHP scripts called by a web server run as a completely different user than you, with completely different permissions.

Do not use fopen/fclose for device files. They are buffered, and overkill besides. Use open/close/read/write calls.
